Third chamber, in fire ant.
What is the sequence you need to perform
I seemed to have tried most things but nothing will make the grey blocks fall down.
Please some help

We have a map of the whole game thanks to Norbert Kintzler. Everything you need to know and more is on it 
But if you don't want to spoil the rest of the game then here is the solution for camber 3, Part 4 is the one your after:
1. Get Key
2. Get Mushroom
3. Open Barrier
4. Run to the left with fire pressed, ladder will break and bricks will drop and dig a hole in the walls.
5. Exit

Totally followed your instructions, and nothing at all.
When you say press fire, is there an actual fire button.
Cos i have been using I,J,K,L and thats it
I am using WINEmu and can't seem to set the controls, although the option is there.

I also found that room was difficult. Fire button is the SPACE key in this game. So once you get the objects in that room and go to that ladder, just move around and press space, it should work.

If using joystick controls, it is indeed the fire button. I never played the game with keyboard so I did not know what is the keyboard replacement for the joystick fire...

| Small correction
About the map: in room two, if you just to to wall (marked as 7 on the map) after picking up the objects, the fly will follow you, you don't need to press the fire button. (That's why Shindig bumped into this problem in room 3, that's the first room you need to use fire.)
